This study was aimed to detect and identify fish betanodavirus, the causative agent of VNN, using nested RT-PCR from brain tissue of Caspian Sea Mullet. Forty samples of subclinical golden grey mullet fish (range: 50-250 gr) were collected from the coastal areas of Caspian Sea in 2016. The target organ (brain) was excised and the reverse transcription (cDNA synthesis) was carried out followed by the RNA extraction. In the present study, detecting of the virus was performed with Nested RT-PCR using two pairs of primers F2/R3 and NF2/NR3 in two steps. After optimization of PCR, all of the forty specimens were negative by the first-step PCR (RT-PCR) while 20 out of 40 grey mullet were positive using Nested RT-PCR (second-step PCR) and a strong band of the expected size (300 bp) was clearly observed. Therefore, it seems that Nested RT-PCR is a method of choice for rapid detection of the low level of infection and in asymptomatic carrier fish. In addition, the result of sequencing revealed that the virus belonged to the RGNNV genotype.}, keywords = {Viral Nervous Necrosis,Golden grey mullet,Nested RT-PCR,Sequencing}, title_fa = {معرفی روش مولکولی PCR آشیانه ای (Nested RT-PCR) اصلاح شده جدید جهت تشخیص سریع بیماری نکروز عصبی ویروسی (Viral Nervous Necrosis)}, abstract_fa = {بیماری نکروز عصبی ویروسی، یکی از مهمترین بیماری­های ویروسی با گسترش جهانی در ماهیان دریایی و پرورشی دنیا محسوب می­شود و تا به امروز بیش از 70 گونه حساس ماهی به این بیماری شناسایی شده است. هدف از این پژوهش ردیابی و شناسایی سریع ویروس مذکور با استفاده از روش Nested RT-PCR از بافت مغز کفال ماهیان دریای خزر بود. در طول فصل صید در سال 1395 تعداد 40 قطعه کفال طلایی (Lizaaurata) در محدوده وزنی 50  تا 250 گرم با علائم مزمن بیماری از سواحل دریای خزر جمع آوری شدند و پس از جداسازی بافت مغز و استخراج RNA ویروس، با استفاده از روش ترانس کریپتاز معکوس، سنتز cDNA از روی RNA ژنوم ویروس صورت گرفت. سپس با روش Nested RT-PCR و با استفاده از دو جفت آغازگر اختصاصی F2/R3 و NF2/NR3 طی دو مرحله، قطعه مورد نظر تکثیر و توالی یابی شد. پس از بهینه سازی شرایط، واکنش PCR با استفاده از نمونه­ی کنترل مثبت، در PCR مرحله ی اول (RT-PCR) در هیچ یک از نمونه­های مشکوک کفال ماهیان باند مورد نظر مشاهده نشد. اما در دور دوم PCR (Nested RT-PCR ) در 20 مورد از 40 نمونه کفال ماهیان یک باند واضح bp 300 مشاهده شد. براساس نتایج بنظر می­رسد که Nested RT-PCR می­تواند به عنوان یک روش مناسب برای تشخیص سریع سطوح پایین آلودگی و همچنین یک روش مناسب و کاربردی برای شناسایی ماهیان حامل آلوده و به ظاهر سالم در نظر گرفته شود. همچنین نتایج حاصل از توالی یابی نشان داد که ویروس مورد نظر در کفال ماهیان دریای خزر، متعلق به ژنوتیپ RGNNV می باشد.}, keywords_fa = {نکروز عصبی ویروسی,ماهی کفال,Nested RT-PCR,توالی‌یابی}, url = {https://isfj.areeo.ac.ir/article_118421.html}, eprint = {https://isfj.areeo.ac.ir/article_118421_d5e124a7294de9541cc3d7bfd74e064e.pdf} }
