بررسی اثر سیتوتوکسیک و ضد سرطانی دیواره بدن خیار دریایی گونه Holothuria leucospilota در شرایط آزمایشگاهی

در سال‌های اخیر تلاش­ها به‌منظور یافتن مواد زیست فعال از جانداران به‌خصوص جانوران دریایی افزایش‌یافته است. در طی این پژوهش اثر ضد سرطانی و سیتوتوکسیک دیواره بدن خیار دریایی گونه Holothuria leucospilota مورد بررسی قرار گرفت. برای این منظور خیار‌های دریایی از عمق 10 الی 30 متری اطراف جزیره لارک جمع‌آوری و عصاره ­گیری از آن‌ها با استفاده از حلال متانول و دی اتیل اتر انجام شد. عصاره‌های به‌دست‌آمده پس از تغلیظ به‌ وسیله دستگاه روتاری در دمای 40 درجه سانتی‌گراد، به‌وسیله دستگاه وکیوم فریز درایر به‌صورت پودر خشک درآمد و مورد بررسی قرار گرفت. به‌منظور بررسی اثر سیتوتوکسیک و ضد سرطانی دیواره بدن خیار دریایی از روش رنگ سنجی XTT استفاده شد. نتایج آزمایش نشان داد که عصاره‌ متانولی استخراج‌ شده در غلظت‌های 100 و 500 میکروگرم بر میلی‌لیتر و عصاره دی اتیل اتری در غلظت 500 میکروگرم بر میلی‌لیتر توان ممانعت از تکثیر سلول‌های سرطانی اپیدرمویید دهان انسان در محیط آزمایشگاهی را داشتند. همچنین این عصاره­ها دارای اثر سیتوتوکسیکی قویی بر روی سلول‌های نرمال بودند. درمجموع دیواره بدن خیار دریایی دارای اثر سیتوتکسیک بسیار قوی بود و می­توان از آن به عنوان یک ماده سیتوتوکسیک یاد کرد. ولی این عصاره­ها ارزش درمانی قابل توجه­ای به منظور درمان سلول­های سرطان دهانی از خود نشان ندادند.

In recent years efforts to find bioactive compounds from live organisms especially marine animals have been increased. In the present study, the anticancer and cytotoxic effects of sea cucumber body walls (Holothuria leucospilota) were investigated. For this purpose, sea cucumbers were collected from Larak Island at depths of 10 to 30 m and extraction process was done with methanol and diethyl ether solvent which then concentrated by rotary evaporator (40℃) following lyophilization with vacuum freeze dryer. XTT method was used to investigate anticancer and cytotoxic effects of body wall extracts. The results showed that the methanolic extract could prevent proliferation of human oral epidermoid carcinoma cells (KB) at concentrations of 100 and 500 μg/ml. The diethyl etheric extract also could prevent proliferation of KB at 500 μg/ml concentration. Overall result showed that sea cucumber body wall had a strong cytotoxic effect on normal cell line (Human embryonic kidney cell [HEK]) which can be used as potent cytotoxic material. However these extracts did not show significant therapeutic value against KB cells.
